چکیده

It has been postulated that cancer chemo therapy utilizing metabolic antagonists may be based on quantitative differences between normal and tumor tissue. In general, neoplasms have con siderably lower enzyme and vitamin concentra tions than most normal tissues, and, in some cases, the observed levels are lower in the cancer cell than in any normal tissue. Furthermore, it is known that inhibition by an antimetabolite de pends upon the concentration ratio of the metabo lite, or susceptible enzyme, and antagonist. Thus, it is reasoned that an enzyme in low concentration in cancer tissue may be blocked by means of an antagonist, while the activity of the same enzyme in nonmalignant cells would be only minimally depressed owing to its higher concentration. This concept has been previously recorded. Boyland (6) selected a catalase inhibitor, a-nitroso-/3-naphthosol for chemotherapeutic trial, reasoning that the significantly lower enzyme concentration in certain tumor tissues should per mit inhibition of tumor growth with doses of this agent tolerated by normal tissues. Ackermann and Potter (1) stated that ". . . it should be possible to

The effect of Vitamin B6 on chemotherapy induced nausea and vomiting in pediatric cancer

Background: Nausea and vomiting are the common side-effects of chemotherapy in children with malignancy. In this study, the effectiveness of vitamin B6 in reducing the chemo-induced nausea and vomiting (CINV) in children was tested. Material and methods: A triple-blind clinical trials was performed on 100 children with malignancy referring to the pediatric clinic of Amir Kabir Hospital, Arak, ...

Association between pre-chemotherapy serum levels of vitamin D and clinicopathologic findings in gastric cancer

Background: To examine serum levels of vitamin D in newly diagnosed gastric cancer (GC) patients compared with normal subjects and any possible association with prognostic variables. Methods: One-hundred subjects (50 GC and 50 controls) were enrolled and serum vitamin D levels were assessed using ELISA. Based on two definitions, vitamin D was classified as a sufficient level (≥30 ng/dL) and op...

A possible basis for the chemotherapy of neo

plastic disease rests in the quantitative differences in the nutrient and coenzyme concentrations be tween the tumor and the host's normal tissues. Various observers have noted that, in general, vitamins tend to be in lower concentration in the malignancy (4, 7, 12, 14—16,18, 19). Systematic measurement of various vitamins and vitamin containing coenzymes has been part of a program in which t...

Quantitative biochemical differences between tumor and host as a basis for cancer chemotherapy. IV. Niacin and 2-ethylamino-1, 3, 4-thiadiazole.

Neoplasms, in general, have considerably lower metabolite and enzyme concentrations than do most normal tissues (1, 5, 6, 8, 10, 12, 14, 15). Since competitive inhibition by an antimetabolite depends upon the concentration ratio of a metab olite and its antagonist, it is reasoned that an antagonist may interfere with some metabolic step to a greater degree in cancer tissue than in those normal ...

Quantitative biochemical differences between tumor and host as a basis for cancer chemotherapy. V. Niacin and 6-aminonicotinamide.

Many metabolites, cofactors, and enzymes have been reported to be in lower concentration in most neoplasms than in the majority of normal tissues of their hosts (8, 11—13,19—@1). These observa tions, together with our growing knowledge of the mechanism of action of antiinetabolites, have given rise to the concept that it may be possible to block metabolic pathways in malignant tissue while ...
